Abstract        : Precision medicine has demonstrated that genetic features of cancer cells, i.e., gene expression, can be used for predicting drug response. Emerging evidence suggests that genedrug connections could be predicted more accurately by simultaneously exploring the cumulative effects of many genes. We developed DeSigN, a web-based tool for predicting drug efficacy against cancer cell lines using gene expression patterns. The algorithm correlates phenotype-specific gene signatures derived from differentially expressed genes with predefined gene expression profiles associated with drug response data (IC50). We and others subsequently demonstrated that DeSigN could robustly predict drugs that could benefit tumour control. For instance, less than 20% of head and neck cancer (HNC) patients, including oral cancer, would respond to three FDA-approved targeted therapies, underscoring the urgent need to develop more effective therapies. Using gene expression signatures derived from oral cancer, DeSigN predicted that bosutinib, an Src/Abl kinase inhibitor, as a sensitive inhibitor for oral squamous cell carcinoma (OSCC) cell lines. As a further confirmation, we demonstrated experimentally that bosutinib has anti-proliferative activity in OSCC cell lines. Here, we showed that DeSigN could be a means to identify efficacious drugs to fight cancers.
